The Division of Hematology and Oncology features a rich history with strengths in basic, clinical and translational research; a dedication to robust educational endeavors; and a commitment to improve the treatment of patients with cancer in Orange County and beyond. Your Role on the Team This position entails providing technical support for projects related to overcoming cancer cell treatment resistance in the Fruehauf lab. Candidate will be responsible for screening small molecules targeting a HIF 1 alpha and HIF 2 using cell models and biochemical techniques. Enzalutamide (Enz) is an effective treatment for castration-resistant and metastatic castration sensitive prostate cancer, but development of resistance is inevitable. Emergence of an alternative splicing variant of the androgen receptor (AR), AR-V7, which lacks the androgen binding and hinge region domains, contributes to Enz resistance in 75% of cases. Nuclear translocation of full-length AR (FL-AR) requires the hinge region. We have found that AR-V7 gains nuclear entry via formation of a heterodimer with HIF 1 alpha, facilitating its nuclear localization, and that blocking HIF translation can restore Enz sensitivity. We have shown that Topotecan blocks HIF1 alpha translation, restoring ENZ activity. We are screening a library of other HIF1 translation inhibitors for potential use in the clinic.
